What exactly are you having problems with in doing this?

From the MapSource help file ...

To create a route using the Route Tool:

Click the Route tool .

Click one or more points on the map in the sequence you want them to appear in the route. If there is not an existing waypoint where you click, a new waypoint is created at that location. NOTE: Waypoints created in this manner do not appear in the Waypoints tab.

After you click all the points you want to include in the route, press Esc on your keyboard or right-click and select Cancel to stop selecting waypoints. The route is created and automatically named using the first and last waypoints. The route appears in the Routes tab on the left side of the screen.

To transfer data from MapSource to a device:

Verify that the device is powered on, properly connected to the computer, and that the device’s transfer mode is set to Garmin (if applicable). Refer to your GPS Owner’s Manual for further instructions.

If the GPS device holds a storage card, verify that the card is inserted correctly in the unit and is not a pre-programmed data card.

Select Send to Device in the Transfer menu.

Choose the device or click Find Device if your device does not appear in the list. When MapSource detects the device, the name of the connected device appears in the Device field.

NOTE: By default, MapSource searches all connected serial ports for your device. If necessary, you can use the Transfer preferences tab to specify a serial port on which to search for a device. See Setting Preferences for more information.

Place a check mark next to the items you want to send to the GPS device.

NOTE: If an option is grayed out, it is not available for your unit, or you have not selected that type of data in MapSource.

Click Send. The data is sent to the GPS device.

I have a 2720

and do this frequently - if you could provide some information about what you are having problems with or don't understand, I'd be glad to walk you through the process - it is not all that difficult.
