I have Garmin Nuvi 350. Is there any where I can set speed alert ? Do I need Custom POI data to do this or this could be done by enabling some feature.

You would need to create a Custom POI file with the points at which you want to be alerted. Then you would set the speed alert either in the file or when you loaded the file with POI Loader.

Take a look at the POI Loader help file under the section titled "Speed and Proximity Alert Information".
