I am a nuvi with my Nuvi 350 and POI's. How does a custom POI interact with the Nuvi when downloaded. Example. I just loaded the Passport America Campground POI. Will a campground show up on a route? Can I type in Passport America and will the Nuvi find the closet one to my current location?

Just trying to find out how these POI's function with the GPS
Press "Where To?", "My Locations", "Custom POIs" (assuming you loaded them correctly to your unit), "Passport America" (or whatever name you have for the file). You will see the 50 closest to your current location. It will show up on your map if it is selected as the destination (at least on my unit c330). Hope this helps...

Do yourself a favor if you are going to download a lot of POI for your GPS and buy a 1 or 2 GB memory card / Stick and save everthing to it

This is up for debate. There has been a lot of discussion on this website that the card is slower. There really is no reason to do this as the nuvi350 has plenty of memory unless you're really going to load it up with millions of POIs (and then, if necessary, you can remove unused voices to get more memory). I put my music MP3s on the card and all of my POIs (I have a lot of them) on the nuvi. Never had a problem.
