About all I have used it for is a larger map. It can be used to download you GPS tracks to see where you have been on the map. I believe it can be used for creating POI's but I have not tried. I did not really see any real use until I did my free map update from Garmin. If you GPS supports it I think you can also use it to create a route and upload it to the GPS.
My 2 cents is...
... I have roadtrip on my Mac, and find it close to useless. It's a poor program with few bells and whistles. The Windows version (Mapsource) is much better, especially if you want to upload to a GPS stuff such as routes for hiking or boating. I run the Windows version under Parallel 4.0 and it works fine.

Update Roadtrip
I'll agree, when it first came out, Roadtrip was spartan. I have used Mapsource for a couple years, and when I purchased a Macbook, installed Roadtrip. It was only after a year of using both, I decided to stick with Roadtrip, to download maps (that were made on Mapsource), and to create routes. I really don't do much beyond route design, and downloading routes, and Roadtrip works fine for that. If you create POI files, perhaps having a PC, with Mapsource installed, is the way to go. for now. love my Mac...
