Is there any way of Programmatically creating garmin routes from poi files or is there an external program that would do this?

Sorry
Your 250 does not do routes.
Nuvi 660 -- and not upgrading it or maps until Garmin fixes long-standing bugs/problems, and get maps to where they are much more current, AND corrected on a more timely basis when advised of mistakes.

Sorry,
I now have a 760. So any ideas?

You can use MapSource to plan routes and send them to your 760. If you don't have it, call Garmin and ask for it. They will likely send it to you at no cost or for a small shipping cost.
(Note that in order for your Custom POI files to show up in MapSource, they need to be GPX file format. Then you can open the files directly in MapSource.)
Or you can use MapQuest online to plan routes and send them to the unit with the Garmin Communicator plug-in. (Won't necessarily use your Custom POIs, though. You would have to search for the locations in MapQuest and enter them in the route.)
Or you can plan them on the unit itself and save them on the unit.
Or you can do a combination of these. You could start a route off the unit, send it to the unit, and then edit it on the unit to include locations in your Custom POI.
.
Note that if you're sending a completed route from MapSource to the 760, you will need to import the route to the GPS using Tools/My Data to be able to use it.
